Abstract: Periodontitis is an inflammatory disease caused by bacterial infection at the surface and inflammatory response against these microbes. Aggregatibacter actinomycetemcomitans (Aa), associated with aggressive periodontitis, commonly produce the cytolethal distending toxin (CDT), a holotoxin composed of 3 subunits, CdtA, CdtB and CdtC. An active CdtB subunit has been reported for inositol polyphosphate 5-phosphatase and DNase activities. This study aimed to observe the CdtB produced by Aa in different media. PCR amplification confirmed that the bacteria culture were Aa. Also the mutation status of both WT and CdtB strain was confirmed by PCR targeting CdtB gene. We then measured the growth and CdtB protein expression of Aa in different medium. Doubling time of Aa in BHI medium was shorter than RPMI medium. CdtB protein expression of Aa was increased in parallel with the growth.
